High Throughput Patient Derived Tumoroids Culture for Functional Precision Radiotherapy
Precision oncology relies on genomic and molecular profiling to identify potentially actionable alterations. However, molecular insight does not accuratelly predict whether a specific tumour will respond to a specific treatment, particularly when sensitivity is influenced by non-genetic cellular states, pathway interactions, and microenvironmental factors. Functional precision oncology has therefore emerged as a complementary approach in which viable patient-derived tumour material is used to test candidate therapies and generate experimentally measured treatment-response profiles with the potential to guide treatment selection more efficiently. Combining advanced microfabrication and patient-derived brain tumour cells, we are developing novel approaches to advance functional precision radiotherapy.
Perfusion Culture of Organotypic Brain Tumour Slices
Perfusion culture of organotypic tumour slices offers significant benefits in cancer research by closely mimicking the in vivo tumour microenvironment. It enhances nutrient and oxygen supply to the slices, promoting cell viability and functionality over extended periods. It allows for more accurate assessment of tumour biology, drug responses, and intercellular interactions. Our approach relies on a pumpless perfusion device designed to better preserve the viability brain tumour tissues ex vivo - a key requirement towards more predictive assessment of therapeutic efficacy.
Long Term Culture of Paediatric Low Grade Glioma
Paediatric low-grade gliomas (pLGGs) are the most common brain tumours in children. Although long-term survival rates exceed 90%, significant treatment associated life-long morbidity emphasizes the need for better balancing the oncological outcome with quality of life of survivors.
